I have a base defense with two entrances on each side of the base. One is open from the start while the other is barricaded off at first. I want all the units to attack move to through the primary entrance at first and latter break the barricade and go through both.
If I put destructible barricades on the ramps the zerglings clump up around it. If I put no pathing on the ground the units will all go through the right entrance, but I don't know if I can turn that off with a trigger.
If this is about killing a pathing blocker you could use a Dynamic Pathing Blocker, it's in the units layer somewhere. You can easily add the following action when you want it removed:
(Unit type of (Triggering unit)) == Space Platform DestructibleSpacePlatformBarrier (Unnamed) - Destructible
Unit - Remove (Closest unit to (Position of (Triggering unit)) in (Dynamic Pathing Blocker 1x1 units in (Entire map) owned by player Any Player matching No Unit Filter, with at most Any Amount)) from the game

You could try placing some Pathing Blocker doodads and then destroy them with triggers.
