They can be .tga also. I use .tga to avoid surplus format transformations.
After you import (F9) the images, you need to save the map, and then you can create buttons (under buttons tab), then pick your image for the respective field. And then you can use your button by picking it from the list when you, for example, create a button for command card.
Having graphical stuff in assets/textures is only important for textures when a model expects to find them there.
Usually it's just convenient to keep imported textures in a distinct folder for easier access. Because when you need to pick a pic, you must remember its name or you will never find it amongst assets/textures crap.

You guys have created nice icon packs here but I don't know yet how I can add custom buttons for my mod. Any tutorials about this?

Also, what programs can open .dds files? At least GIMP can't open.

You import files using the Import Manager.

So quick tutorial for other newbies:
1. Press F9
2. Import files (select icons from pack you downloaded)
3. Save
@Terhonator: You also need to check if they are in the correct folder. For any icons or textures the path is Assets/Textures/
OK. Thanks I had these in root and it worked fine actually.
To move files right click over file and select "Move files" and to new path type "Assets/Textures"
